ATP detection on cell surfaces
For the quantification of ATP-concentrations on cell surfaces, ATP-sensitive organo-Zn-complexes will be prepared.In this context, three major criteria are: i) a selective binding of ATP, ii) a sufficient sensitivity towards ATP, and iii) the option to anchor the sensors to cell surfaces by covalent linkages with the aim of measuring local, extracellular ATP-concentrations. Apart from lipophilic anchor residues which spontaneously integrate into the membrane, sensors with anchors comprising azide or alkyne moieties will be synthesized in order to covalently attach these to alkyne or azide modified proteins integrated into the membrane by using “Click”-chemistry.
