B3: Aufklärung molekularer Erkennungsmotive in Glycosyltransferase/Ligand-Komplexen

Keywords: NMR, Glycosyltransferases, Bioactive Conformations, Carbohydrate-Protein Interactions, Surface Plasmon Resonance

The mechanism of glycosyltransferases is investigated utilizing different biophysical techniques. The most important tool is high-field NMR spectroscopy that is employed to define the bioactive conformations of donor- and acceptor-ligands as well as their binding epitopes.
Using uniform and amino acid selective isotope labeling with 15N, 13C, and 2H we also investigate the protein conformation and the topology of the binding site.
Our current interest is focussed on galactosyl- and sialyltransferases.
