A5: Enzymatische Glycosylierungen und AFM-Untersuchungen

Keywords: Chemoenzymatic synthesis, glycosyltransferases, transsialidase, transglycosylation, functionalized NDP-sugar substrates

This project is concerned with chemical as well as chemoenzymatic syntheses of complex bioactive glycostructures. Transsialidase (Trypanosoma cruzi) was expressed, and one focus is its use for preparative sialylation employing novel donor substrates and various disaccharide acceptors. Another question deals with a combinatorial use of hydrolase- catalyzed transglycosylations. Further, L-galacto-syltransferase reactions for the synthesis of H-antigen type oligosaccharides are studied. Finally, pathways to obtain membrane-permeant as well as certain affinity labeled NDP- sugar donor substrates will be elaborated.
